Usage and Biology
We placed a reporter gene, sfGFP (BBa_K1624004), downstream of the pCoof-RBS in the pCDFDuet-1 plasmid. We transformed this plasmid along with another plasmid containing a constitutively produced transcription activator CooA into BL21(DE3) cells. We used cells without the modified plasmid as our negative control, and cells containing plasmids with T7-regulated sfGFP as our positive control. We tested the constructs at 0, 50, 100, 200 and 500 uM CORM-2 (carbon monoxide releasing molecule). pCoof regulated sfGFP did not show a significant correlation between gene expression and CO presence.
